The BSK-H Medium is a high quality nutrient liquid that is used for the reliable cultivation of Borrelia spec. This medium has a complex formulation with an extraordinary high content of proteins and peptides, it is especially rich with nucleosides, glucose, and contains high concentrations of vitamins. This medium contains N-Acetyle-D Glucosamine that is an essential element for bacterial poptideoglycan.
To produce the BSK-H medium, only a selected few reagents of the highest quality are used. There is a special galenic as well as a gentle and controlled process guarantee a product with the highest quality and reliable lot to lot consistency, stability, purity, and cell growth. There is a high amount of HEPES which allows for stable buffer capacity for a long time. Concentration levels of CO2 and pH value need to be monitored carefully as due to the metabolization of glucose in long-term cultures, lactic acid accumulation occurs that can result in a reduction of the pH-value to more than 1 pH per unit. The pH value is 7.6±0.2 at an osmolality of 420±20 mOsm/kg H2O.
This ready-to-use medium must be supplemented with 3-8% rabbit serum prior to inoculation. The instructions of storage temperature and stability are according to the lot-specific label. The BSK-H medium that can be widely used for the cultivation of spirochetes, especially for B. Burgdorferi and B. Hermsii. Small amounts of organisms are sufficient for the inoculation but there is no general recommendation for the inoculum number as research varies too much. The generation time lies between 11 and 18 hours so that in 7 to 9 days 0.5-4.0x108 cells/ml can be obtained. Optimal incubation temperature is between 30°C and 37°C.
This semi-defined medium for Trypanosoma Brucei was developed by Brun and Jenni and was successfully used to culture various species of Trypanosome. It is available in both liquid and powder form.
